Yes, all bosses can be re-fought by re-entering their dungeon after a 24-hour (in-game) respawn timer. Each kill has a base unique drop rate that scales with difficulty — Normal mode is roughly 15 percent, Hard is 30 percent, and Nightmare is 50 percent. The Elder Vampire Lord has a guaranteed legendary drop on the first kill of each real-world week. Farming bosses is the primary endgame loop for perfecting your gear.
Do I need a different build for each boss?
A well-rounded build can handle the first three bosses without respecialization, but the Plague Sovereign and Elder Vampire Lord strongly reward specialized builds. The Plague Sovereign is nearly impossible without fire damage to clear poison clouds.
